Tirzepatide is a synthetic peptide that acts as a dual agonist at the glucose-dependent insulinotropic polypeptide (GIP) and glucagon-like peptide-1 (GLP-1) receptors. The molecule contains 39 amino acids and features a C20 fatty diacid moiety attached via a linker, which promotes albumin binding and extends its circulating half-life. Its sequence incorporates non-natural amino acids and modifications that reduce susceptibility to degradation by dipeptidyl peptidase-4. This dual receptor activity distinguishes it from selective GLP-1 receptor agonists.
The GIP receptor is expressed in pancreatic islets, adipose tissue, and the central nervous system, while GLP-1 receptors are found in pancreatic islets, the gastrointestinal tract, and the brain. Activation of both receptors can enhance glucose-dependent insulin secretion and reduce glucagon release. The relative contribution of each receptor to the overall pharmacological effect remains an area of ongoing investigation. Preclinical studies suggest that GIP receptor agonism may modulate appetite and energy balance, but the precise mechanisms in humans are not fully established.
In clinical research, tirzepatide has been studied in randomized controlled trials for glycemic control and body weight reduction. These trials typically measure changes in hemoglobin A1c and body weight over periods of several months. The drug is administered by subcutaneous injection, and its pharmacokinetic profile supports once-weekly dosing. Post-marketing surveillance continues to evaluate long-term outcomes and rare adverse events.
Identity and purity are usually established with reversed-phase high-performance liquid chromatography for the main peak and with mass spectrometry for the observed molecular mass. Peptide mapping after enzymatic digestion confirms the primary sequence, while amino acid analysis provides a quantitative composition check. Size-exclusion chromatography and ion-exchange chromatography are used to look for aggregates and charge variants. Water content, residual solvents, and counter-ion content are measured separately, since a lyophilised powder is often reported on an as-is basis unless a correction is applied.
Research-grade material circulates through suppliers that differ widely in documentation and testing practice, so a certificate of analysis is a starting point rather than proof of quality. Independent verification typically repeats chromatographic purity and mass confirmation on the received lot, and compares results against a retained reference standard. Regulatory status varies by jurisdiction, and a substance cleared as a medicine is not interchangeable with a research chemical of the same name. Open questions include how closely non-pharmaceutical lots match approved material in impurity profile and in aggregate content.
Solid tirzepatide is handled as a lyophilised, hygroscopic peptide powder that should be kept desiccated, protected from light, and stored frozen, typically at or below minus twenty degrees Celsius for long-term retention. Material left at ambient temperature for extended periods can take up moisture, which promotes aggregation and deamidation. Commercial liquid presentations are kept refrigerated between two and eight degrees Celsius and are not frozen. Reconstituted laboratory solutions are generally held cold and used within a short window because hydrolysis and oxidation continue slowly in solution.

Structure-activity work shows that fatty acid length, linker chemistry and the position of acylation all influence albumin affinity and receptor potency. Plasma protein binding exceeds 99 percent, which restricts distribution and slows renal clearance. Degradation proceeds largely through general proteolysis and fatty acid oxidation rather than cytochrome P450 metabolism, so exposure to common oxidative drug interactions is limited. Whether these clearance routes vary meaningfully between individuals is not fully established.
The molecule is a synthetic 39-amino-acid peptide whose backbone derives from the sequence of human glucose-dependent insulinotropic polypeptide, with several substitutions that raise metabolic stability and shift receptor preference. A C20 fatty diacid is attached through a short linker to a lysine side chain, a modification that increases binding to serum albumin. The reported monoisotopic mass is approximately 4813 Da. Near neutral pH the peptide carries a net negative charge, and the lipid tail makes the molecule markedly more hydrophobic than the unmodified parent sequence.
Dual agonism at the GIP and GLP-1 receptors underlies the observed pharmacology. Activation of GLP-1 receptors raises glucose-dependent insulin release, lowers glucagon secretion, slows gastric emptying and reduces appetite. GIP receptor activation contributes additional effects on adipose tissue and on energy balance, and the combined action on appetite appears larger than either pathway alone in animal models. Signalling bias and the relative contribution of each receptor arm to weight-related effects remain areas of active investigation.
The peptide backbone contains 39 amino acids and includes alpha-aminoisobutyric acid residues, which are not among the standard proteinogenic set. A C20 fatty diacid moiety is attached through a linker, allowing the compound to bind serum albumin and extend its circulation time. This albumin binding is the main reason the molecule supports once-weekly administration rather than more frequent dosing. The measured molecular mass is approximately 4,813 daltons, placing it firmly in the peptide rather than small-molecule class.
Tirzepatide is a synthetic peptide that activates both the glucose-dependent insulinotropic polypeptide (GIP) and glucagon-like peptide-1 (GLP-1) receptors. This dual agonist profile distinguishes it from earlier incretin-based compounds that act on a single receptor. The molecule was engineered from the native GIP sequence and carries several non-natural residues that slow enzymatic breakdown. Researchers designed it to combine the insulinotropic effects of GIP signaling with the appetite and gastric-emptying effects associated with GLP-1 activation.
People convicted of murder, drug trafficking, corruption or human rights violations will not qualify under the amnesty law. The measure was long requested by the US-backed Venezuelan opposition. Opposition leader María Corina Machado said that the bill was not taken "voluntarily, but rather in response to pressure from the US government". Rodríguez also announced the shutdown of El Helicoide prison, which she said would be transformed into a cultural and sport center. Independent organizations have documented many cases of torture and human rights violation in El Helicoide. Family of some of the prisoners gathered outside El Helicoide chanting "Freedom!". Venezuelan opposition politician and ally of Machado, Juan Pablo Guanipa, was released from prison on 8 February but taken by unknown armed groups hours later; by 10 February, his family confirmed he was under house arrest in Maracaibo, calling the measure unjust detention. On 19 February, the amnesty law was approved by the National Assembly of Venezuela. Jorge Rodríguez said that over 1557 request liberty a day after. Juan Pablo Guanipa was also released from house arrest. During the 2026 State of the Union Address, Trump invited Venezuelan politician Enrique Márquez, who was imprisoned in 2025 and was released in early January. Trump said "Since the raid we have worked with the new leadership and they have ordered the closure of that vile prison and released hundreds of political prisoners already, with more to come."
=== Career === In 1966, Kenyon started as an assistant professor of biology at San Francisco State University and became emeritus in 2001. In 1969, Kenyon co-authored Biochemical Predestination with Gary Steinman. Chemist Stephen Berry explained Kenyon's and Steinman's theory as "describing the following causal chain: the properties of the chemical elements dictate the types of monomers that can be formed in prebiotic syntheses, which then dictate the properties of the occurring polymers, which finally dictate the properties of the first eobionts and all succeeding cells." Kenyon's work was about virus production. During the 1969–1970 academic year he was "on a fellowship at the Graduate Theological Union in Berkeley, where he reviewed the contemporary literature on the relationship of science and religion." Then in 1974, he was a visiting scholar to Trinity College, Oxford. Kenyon states that his views changed around 1976 after exposure to the work of young Earth creationists:
=== Detection in body fluids === Buprenorphine and norbuprenorphine may be quantified in blood or urine to monitor use or non-medical recreational use, confirm a diagnosis of poisoning, or assist in a medicolegal investigation. A significant overlap of drug concentrations exists in body fluids within the possible spectrum of physiological reactions ranging from asymptomatic to comatose. Therefore, knowing both the route of administration of the drug and the level of tolerance to opioids of the individual is critical when results are interpreted.
=== ES-SCLC === In ES-SCLC, platinum-based combination chemotherapy is the standard of care. Combination chemotherapy consists of a wide variety of agents, including cisplatin, cyclophosphamide, vincristine and carboplatin. Response rates are high even in extensive disease, with between 15% and 30% of subjects having a complete response to a combination chemotherapy, and the vast majority having at least some objective response. Responses in ES-SCLC are often of short duration, and the evidence surrounding the risk of treatment compared to the potential benefit of chemotherapy for people who have extensive SCLC is not clear.

== Extracellular NAMPT == Extracellular NAMPT (eNAMPT) is functionally different from intracellular NAMPT (iNAMPT), and less well understood (which is why the enzyme has been given so many names: NAMPT, PBEF and visfatin). iNAMPT is secreted by many cell types (nobably adipocytes) to become eNAMPT. The sirtuin 1 (SIRT1) enzyme is required for eNAMPT secretion from adipose tissue. eNAMPT may act more as a cytokine, although its receptor (possibly TLR4) has not been proven. It has been demonstrated that eNAMPT could bind to and activate TLR4. eNAMPT can exist as a dimer or as a monomer, but is normally a circulating dimer. As a monomer, eNAMPT has pro-inflammatory effects that are independent of NAD+, whereas the dimeric form of eNAMPT protects against these effects. eNAMPT/PBEF/visfatin was originally cloned as a putative cytokine shown to enhance the maturation of B cell precursors in the presence of Interleukin-7 (IL-7) and stem cell factor, it was therefore named "pre-B cell colony-enhancing factor" (PBEF). When the gene encoding the bacterial nicotinamide phosphoribosyltransferase (nadV) was first isolated in Haemophilus ducreyi, it was found to exhibit significant homology to the mammalian PBEF gene. Rongvaux et al. demonstrated genetically that the mouse PBEF gene conferred Nampt enzymatic activity and NAD-independent growth to bacteria lacking nadV. Revollo et al. determined biochemically that the mouse PBEF gene product encodes an eNAMPT enzyme, capable of modulating intracellular NAD levels. Others have since confirmed these findings.
== Mechanism == Ammonium sulfate, as well as other neutral salts, will stabilize proteins by preferential solvation. Proteins are usually stored in ammonium sulfate because it inhibits bacterial growth. With the addition of ammonium sulfate, proteins unfolded by denaturants can be pushed into their native conformations. This can be seen with the folding of recombinant proteins. The solubility of proteins varies according to the ionic strength of the solution, thus according to the salt concentration. At low ion concentrations (less than 0.5 mol/L), the solubility of proteins increases with increasing salt concentration, an effect termed "salting in". As the salt concentration is further increased, the solubility of the protein begins to decrease. At a sufficiently high ionic strength, the protein will precipitate out of the solution, an effect termed "salting out". When the ammonium (NH+4) and sulfate (SO2−4) ions are within the aqueous solution they are attracted to the opposite charges evident on the compound that is being purified. This attraction of opposite charges prevents the water molecules from interacting with the compound being purified, leading to the precipitation or "salting out". Proteins differ markedly in their solubilities at high ionic strength, therefore, "salting out" is a very useful procedure to assist in the purification of the desired protein. Ammonium sulfate is commonly used for precipitation because of its high solubility, additionally, it forms two ions high in the Hofmeister series.
MD simulation of the full satellite tobacco mosaic virus (STMV) (2006, Size: 1 million atoms, Simulation time: 50 ns, program: NAMD) This virus is a small, icosahedral plant virus that worsens the symptoms of infection by Tobacco Mosaic Virus (TMV). Molecular dynamics simulations were used to probe the mechanisms of viral assembly. The entire STMV particle consists of 60 identical copies of one protein that make up the viral capsid (coating), and a 1063 nucleotide single stranded RNA genome. One key finding is that the capsid is very unstable when there is no RNA inside. The simulation would take one 2006 desktop computer around 35 years to complete. It was thus done in many processors in parallel with continuous communication between them. Folding simulations of the Villin Headpiece in all-atom detail (2006, Size: 20,000 atoms; Simulation time: 500 μs= 500,000 ns, Program: Folding@home) This simulation was run in 200,000 CPU's of participating personal computers around the world. These computers had the Folding@home program installed, a large-scale distributed computing effort coordinated by Vijay Pande at Stanford University. The kinetic properties of the Villin Headpiece protein were probed by using many independent, short trajectories run by CPU's without continuous real-time communication. One method employed was the Pfold value analysis, which measures the probability of folding before unfolding of a specific starting conformation. Pfold gives information about transition state structures and an ordering of conformations along the folding pathway.
== Military == The Armed Forces of Serbia and Montenegro (Serbian: Војска Србије и Црне Горе/Vojska Srbije i Crne Gore, ВСЦГ/VSCG), previously known as Army of Yugoslavia (Serbian: Војска Југославије/Vojska Jugoslavije, ВЈ/VJ) included ground forces with internal and border troops, naval forces, air and air defense forces, and civil defense. It was established from the remnants of the Yugoslav People's Army (JNA), the military of the SFR Yugoslavia. Several Bosnian Serb units of the VJ were transferred over to the Republika Srpska, during the course of the Bosnian War, leaving only units directly from Serbia and Montenegro in the armed forces. The VJ saw military action during the Yugoslav Wars, including the Siege of Dubrovnik, as well as the Kosovo War, and played combat roles during ethnic insurgencies. Following the Kosovo War, the VJ was forced to evacuate Kosovo, and in 2003 it was renamed the ''Armed Forces of Serbia and Montenegro.'' Following the dissolution of the Union between Serbia and Montenegro, units from each army were assigned to the independent republics of Serbia and Montenegro, as recruitment in the army was on a local, rather than Federal, level. Montenegro inherited the small navy of the FR Yugoslavia, due to Serbia being landlocked.

It activates both GIP and GLP-1 receptors. This dual action differentiates it from selective GLP-1 agonists.
It is given as a subcutaneous injection. Its long half-life supports weekly dosing.
No, it is synthetic. It contains non-natural amino acids and a fatty acid modification.
It is normally kept frozen, desiccated, and away from light, with brief warming to room temperature before opening to limit condensation. Repeated freeze-thaw cycles are avoided because they stress the peptide. Once in solution, the material is held cold and used promptly.
