Tirzepatide is a synthetic linear peptide of 39 amino acids that acts as a dual agonist at the glucose-dependent insulinotropic polypeptide (GIP) and glucagon-like peptide-1 (GLP-1) receptors. Its sequence derives from native GIP but incorporates non-natural residues and a C20 fatty diacid moiety linked to a lysine side chain. The lipophilic chain promotes albumin binding, which slows renal clearance and extends circulation time. The unmodified peptide has a molecular formula of C225H348N48O68 and a molecular mass near 4,813 daltons.
Receptor activation by tirzepatide raises intracellular cyclic AMP through Gs-coupled signalling at both targets. At the GLP-1 receptor the downstream effect includes glucose-dependent insulin release, suppressed glucagon secretion, delayed gastric emptying, and reduced appetite signalling in the hypothalamus. GIP receptor engagement adds insulinotropic activity and appears to influence lipid handling in adipose tissue. Because both receptors are stimulated at the same time, the pharmacological profile differs from that of selective GLP-1 receptor agonists, and the relative contribution of each arm remains an area of active investigation.
Bulk peptide material is normally characterised by reversed-phase high-performance liquid chromatography, which separates the target sequence from truncation products and other closely related impurities. Ultraviolet detection near 214 nanometres is common because the peptide backbone absorbs in that region. Mass spectrometry, usually electrospray ionisation coupled to a mass analyser, is used to confirm the molecular mass. Because the molecule carries a lipophilic side chain, gradient methods often need a relatively high organic modifier fraction to elute it within a practical retention window.
Like most synthetic peptides of this size, the material is commonly supplied as a lyophilised powder that appears white to off-white. It dissolves in aqueous buffers and in mixtures of water with a small proportion of organic solvent, though the fatty acid portion reduces solubility in pure water relative to short peptides. Hygroscopic behaviour is reported for many peptide powders, so weighing is usually performed quickly and under controlled humidity. Working solutions are typically prepared fresh and kept cold.

Tirzepatide is a synthetic peptide of 39 amino acids that carries a C20 fatty diacid side chain attached through a linker. Its molecular formula is C225H348N48O68, and its molecular weight is about 4813 daltons. The compound belongs to the incretin mimetic class and is administered by subcutaneous injection. The fatty acid chain promotes binding to serum albumin, which slows renal clearance and extends the circulation time of the molecule. It was identified during screening of sequences derived from glucose-dependent insulinotropic polypeptide.
Tirzepatide activates both the glucose-dependent insulinotropic polypeptide receptor and the glucagon-like peptide-1 receptor, making it a dual agonist rather than a selective agent. Engagement of the GLP-1 receptor is linked to glucose-dependent insulin release, slower gastric emptying, and reduced appetite signalling. The relative contribution of the GIP arm remains an active research question; proposed roles include improved insulin sensitivity and altered adipose tissue handling. Receptor occupancy studies suggest the molecule interacts with both targets at circulating concentrations achieved during therapy.
Identity and purity of tirzepatide are assessed mainly by reversed-phase high-performance liquid chromatography with ultraviolet detection, often paired with mass spectrometry. Because the molecule carries several modifications, gradient conditions are adjusted to resolve the intact peptide from deamidation and oxidation products. Enzymatic digestion followed by peptide mapping confirms the primary sequence and locates specific modifications. Quantitation in biological matrices typically uses liquid chromatography with tandem mass spectrometry after solid-phase extraction. Immunoassays are used less often, since antibody cross-reactivity with closely related peptides can bias results.
The peptide shares degradation routes common to modified peptides: deamidation of asparagine and glutamine residues, oxidation of methionine, and backbone hydrolysis under extreme pH. Lyophilized material is generally more stable than a solution, and residual water content directly affects the rate of hydrolysis. In liquid form, aggregation and visible particles can appear after agitation or repeated freeze-thaw cycles. Stability studies therefore track monomer content, aggregate content, and potency over months under defined temperature and humidity.
=== World Health Organization guidelines === The World Health Organization (WHO) published a guideline on protecting workers from potential risk of manufactured nanomaterials at the end of 2017. WHO used a precautionary approach as one of its guiding principles. This means that exposure has to be reduced, despite uncertainty about the adverse health effects, when there are reasonable indications to do so. This is highlighted by recent scientific studies that demonstrate a capability of nanoparticles to cross cell barriers and interact with cellular structures. In addition, the hierarchy of controls was an important guiding principle. This means that when there is a choice between control measures, those measures that are closer to the root of the problem should always be preferred over measures that put a greater burden on workers, such as the use of personal protective equipment (PPE). WHO commissioned systematic reviews for all important issues to assess the current state of the science and to inform the recommendations according to the process set out in the WHO Handbook for guideline development. The recommendations were rated as "strong" or "conditional" depending on the quality of the scientific evidence, values and preferences, and costs related to the recommendation. The WHO guidelines contain the following recommendations for safe handling of manufactured nanomaterials (MNMs) A. Assess health hazards of MNMs
=== Animal rights === In 2005, 2008, and 2010 PETA threatened to submit a shareholder resolution to improve animal welfare standards in the company. In 2005, PETA attempted to stop five specific animal experiments that it deemed "crude and cruel". In 2008, PETA protested the outsourcing of animal testing to countries with lax animal welfare laws, such as China. In 2010, PETA attempted to stop Medtronic's reported use of live animals in testing and training. In response, Medtronic conducted a feasibility study that found that banning the use of live animals was impractical. As of 2015 Medtronic continued to use live animals for testing and training but stated that it would look for alternatives in the future. In each case, PETA withdrew its shareholder resolution after talks with Medtronic's leadership.
== Predisposing factors == Aortic dissection is associated with hypertension (high blood pressure) and many connective tissue disorders. Vasculitis (inflammation of an artery) is rarely associated with aortic dissection. It can also be the result of chest trauma. About 72 to 80% of individuals who present with an aortic dissection have a previous history of hypertension. Use of stimulants such as cocaine and methamphetamine is also a modifiable risk factor for AD. It can also be caused by smoking. A bicuspid aortic valve (a type of congenital heart disease involving the aortic valve) is found in 7–14% of individuals who have an aortic dissection. These individuals are prone to dissection in the ascending aorta. The risk of dissection in individuals with bicuspid aortic valves is not associated with the degree of stenosis of the valve. Connective tissue disorders such as Marfan syndrome, Ehlers–Danlos syndrome, and Loeys–Dietz syndrome increase the risk of aortic dissection. Similarly, vasculitides such as Takayasu's arteritis, giant cell arteritis, polyarteritis nodosa, and Behçet's disease have been associated with a subsequent aortic dissection. Marfan syndrome is found in 5–9% of individuals who had an aortic dissection. In this subset, the incidence in young individuals is increased. Individuals with Marfan syndrome tend to have aneurysms of the aorta and are more prone to proximal dissections of the aorta. Turner syndrome also increases the risk of aortic dissection, by aortic root dilatation.

The tandem mass spectrometry data on over 960,000 molecular standards (as of October 2025) is provided to facilitate the identification of chemical entities from tandem mass spectrometry experiments. In addition to the identification of known molecules it is also useful for identifying unknowns using its similarity searching/analysis. All tandem mass spectrometry data comes from the experimental analysis of standards at multiple collision energies and in both positive and negative ionization modes.
Among the extreme claims used to market goji berries or its juice, often referred to as a "superfruit", is the unsupported story that a Chinese man named Li Qing Yuen, who was said to have consumed wolfberries daily, lived to the age of 256 years (1677–1933). This claim originated in a 2003 booklet by Mindell, who also claimed goji had anti-cancer properties. The booklet contained false and unverified claims. On 29 May 2009, a class action lawsuit was filed against FreeLife in the United States District Court of Arizona. This lawsuit alleged false claims, misrepresentations, false and deceptive advertising, and other issues regarding FreeLife's Himalayan Goji Juice, GoChi, and TaiSlim products. This lawsuit sought remedies for consumers who had purchased the products over the years. A settlement agreement was reached on 28 April 2010, where FreeLife took steps to ensure that its goji products were not marketed as "unheated" or "raw", and made a contribution to an educational organization. As with many other novel "health" foods and supplements, the lack of clinical evidence and poor quality control in the manufacture of consumer products prevent goji from being clinically recommended or applied.

Vosilasarm, also known by the development codes RAD140 and EP0062 and by the black-market name Testolone or Testalone, is a selective androgen receptor modulator (SARM) which is under development for the treatment of hormone-sensitive breast cancer. It is specifically under development for the treatment of androgen receptor-positive, estrogen receptor-negative, HER2-negative advanced breast cancer. Vosilasarm was also previously under development for the treatment of sarcopenia (age-related muscle atrophy), osteoporosis, and weight loss due to cancer cachexia, but development for these indications was discontinued. The drug is taken by mouth. Side effects of vosilasarm may include vomiting, dehydration, constipation, decreased appetite, weight loss, changes in sex hormone levels, elevated liver enzymes, and liver toxicity. Vosilasarm is a nonsteroidal SARM, acting as an agonist of the androgen receptor (AR), the biological target of androgens and anabolic steroids like testosterone and dihydrotestosterone (DHT). However, it shows dissociation of effect between tissues in preclinical studies, with agonistic and anabolic effects in muscle, agonistic effects in breast, and partially agonistic or antagonistic effects in the prostate gland and seminal vesicles. Vosilasarm was developed in 2010 and was first described in the literature in 2011. It was originally developed by Radius Health and is now under development by Ellipses Pharma.
== In popular culture and other media == During Portal, the player explores areas outside of the test chambers where scrawled messages left by Rattmann and others warn of GLaDOS' deception. In particular, while GLaDOS promises that Chell will receive cake for completing the training courses, the messages alert that this reward does not exist, and that "The cake is a lie". The phrase became an Internet meme, leading to numerous cake-related jokes, as well as its adaption as a term relating to a false promise. When writing Portal 2, Wolpaw stated that they were so sick of cake jokes that they purposely avoided any reference to them, save for one subtle nod. Valve has sold several Portal-based prints, T-shirts, and other memorabilia through its own store, often riding on the popularity of certain memes that the series has created. When first released, both were sold out in under 24 hours. Valve also has partnerships with other vendors for similar merchandise. WizKids has released collectible miniatures of the turrets, cores and companion cube(s) within the game. Minecraft officially features the Portal characters Chell, Atlas, and P-Body as cosmetic skins purchasable from the Minecraft Marketplace for Minecraft Bedrock Edition. The skin pack featuring these characters was originally released as DLC for Minecraft Xbox 360 Edition. Ellen McLain voiced the AI of the Jaeger suite in the 2013 film Pacific Rim, explicitly using the GLaDOS voice in the films trailer. Atlas appears as a player-character in the downloadable content package for Runner2.

It is a dual GIP and GLP-1 receptor agonist, frequently grouped with incretin-based peptide therapeutics. It is a peptide rather than a small molecule and is given by subcutaneous injection.
Selective agents engage only the GLP-1 receptor, whereas tirzepatide activates GIP and GLP-1 receptors simultaneously. This difference in receptor coverage is the principal pharmacological distinction emphasised in comparative reviews.
Downstream signalling is partly characterised, but the quantitative contribution of GIP versus GLP-1 receptor activation to metabolic outcomes is not settled. Review articles commonly flag this as an unresolved question rather than a settled finding.
It separates molecules by hydrophobicity, which is effective for distinguishing an intact peptide from truncated or chemically modified forms. A C18 column with an acidic water-organic mobile phase is a standard configuration.
