==== Pressure ulcers ==== Another leading type of chronic wounds is pressure ulcers, which usually occur in people with conditions such as paralysis that inhibit movement of body parts that are commonly subjected to pressure such as the heels, shoulder blades, and sacrum. Pressure ulcers are caused by ischemia that occurs when pressure on the tissue is greater than the pressure in capillaries, and thus restricts blood flow into the area. At some time after the Norman conquest of England in 1066 a motte-and-bailey was erected on the present site of Castle Park. Bristol was held by Geoffrey de Montbray, Bishop of Countances, one of the knights who accompanied William the Conqueror. William ordered stone castles to be built so it is likely that the first parts of Bristol Castle were built by Geoffrey in his reign. After the Conqueror's death (1087), Geoffrey joined the rebellion against William Rufus. Using Bristol as his headquarters, he burned Bath and ravaged Somerset before submitting to Rufus. He eventually returned to Normandy and died at Coutances in 1093. Rufus created the Honour of Gloucester, which included Bristol, from his mother Queen Matilda's estates and granted it to Robert Fitzhamon. Fitzhamon enlarged and strengthened Bristol castle and in the latter years of the 11th century conquered and subdued much of south and west Wales. His daughter Mabel was married in 1114 to Henry I's bastard son Robert of Caen. Her dowry was a large part of her father's Gloucestershire and Welsh estate and Robert of Caen became the first Earl of Gloucester, c. 1122. He is believed to have been responsible for completing Bristol castle. In 1135 Henry I died and the Earl of Gloucester rallied to the support of his sister Matilda against Stephen of Blois who had seized the throne on Henry's death. Stephen attempted to lay siege to Robert at Bristol in 1138 but gave up the attempt as the castle appeared impregnable.
==== LL Cool J ==== Ice-T had a feud with LL Cool J in the late 1980s and early 1990s. Apparently, this was instigated by LL's claim to be "the baddest rapper in the history of rap itself". Ice-T recorded disses against LL on his 1988 album Power. On the album was the track, "I'm Your Pusher", in which a rap music addict declines to buy an LL Cool J record. In the book Check the Technique: Liner Notes for Hip-Hop Junkies, Ice-T said that the song "Girls L.G.B.N.A.F." was also intended as a diss to LL Cool J, by making a crude song to contrast with the love songs that LL was making at the time. On LL's response, "To da Break of Dawn" in 1990, he dissed Kool Moe Dee (whose feud with LL was far more publicized) as well as MC Hammer. He then devoted the third verse of the song to dissing Ice-T, mocking his rap ability ("take your rhymes around the corner to rap rehab"), his background ("before you rapped, you was a downtown car thief"), and his style ("a brother with a perm deserves to get burned"). He also suggested that the success of Power was due to the appearance of Ice-T's girlfriend Darlene on the album cover. Ice-T appeared to have ignored the insults and he had also defended LL Cool J after his arrest in the song "Freedom of Speech". It has been discovered that the inhibition of PI3Kγ significantly reduces inflammation. PI3Kγ is the intended target molecule in the prevention of inflammation as it is the common signalling effector of many different chemokine and receptor types involved in promoting inflammation. Although PI3Kγ is the intended target there are other isoforms of PI3 which perform different functions from PI3Kγ. Since PI3Kγ is specifically regulated by Gβγ, while other isoforms of PI3 are largely regulated by other molecules, inhibiting Gβγ signalling would provide the desired specificity of a therapeutic agent designed to treat inflammation.
