Dual agonism at the GIP and GLP-1 receptors underlies the observed pharmacology. Activation of GLP-1 receptors raises glucose-dependent insulin release, lowers glucagon secretion, slows gastric emptying and reduces appetite. GIP receptor activation contributes additional effects on adipose tissue and on energy balance, and the combined action on appetite appears larger than either pathway alone in animal models. Signalling bias and the relative contribution of each receptor arm to weight-related effects remain areas of active investigation.
Structure-activity work shows that fatty acid length, linker chemistry and the position of acylation all influence albumin affinity and receptor potency. Plasma protein binding exceeds 99 percent, which restricts distribution and slows renal clearance. Degradation proceeds largely through general proteolysis and fatty acid oxidation rather than cytochrome P450 metabolism, so exposure to common oxidative drug interactions is limited. Whether these clearance routes vary meaningfully between individuals is not fully established.
Structural work on the molecule centers on a C20 fatty diacid moiety attached through a linker to the peptide backbone. This side chain promotes reversible binding to serum albumin, which slows renal clearance and supports a prolonged action profile. The peptide backbone incorporates aminoisobutyric acid substitutions that limit recognition by digestive enzymes. Together these modifications produce a molecule that is stable enough for subcutaneous delivery but still dependent on careful manufacturing control. Analytical characterization of the active pharmaceutical ingredient typically follows the conventions used for other synthetic peptides.
Tirzepatide is a synthetic peptide composed of 39 amino acids. It acts as a dual agonist at two incretin receptors, the glucose-dependent insulinotropic polypeptide receptor and the glucagon-like peptide-1 receptor. The molecule was designed by modifying the native sequence of glucose-dependent insulinotropic polypeptide to improve metabolic stability and extend its circulation time. Its structure includes several non-natural amino acid residues and a fatty acid side chain. These features distinguish it from earlier single-receptor incretin analogs studied in the same period.

The peptide shares degradation routes common to modified peptides: deamidation of asparagine and glutamine residues, oxidation of methionine, and backbone hydrolysis under extreme pH. Lyophilized material is generally more stable than a solution, and residual water content directly affects the rate of hydrolysis. In liquid form, aggregation and visible particles can appear after agitation or repeated freeze-thaw cycles. Stability studies therefore track monomer content, aggregate content, and potency over months under defined temperature and humidity.
Cold-chain handling is standard for formulated product, with dry powder stored frozen and ready-to-use solutions refrigerated. Light exposure is minimized because photodegradation of certain amino acid side chains is possible. Shipping and temperature-excursion studies are used to establish whether short deviations affect quality attributes. Documentation supplied with research material usually includes a certificate of analysis listing purity, identity confirmation, and water or residual solvent content. Users are expected to confirm that material meets the stated specification before use.
Both receptors are class B G protein-coupled receptors that signal largely through Gs-mediated cyclic AMP production. Activation within pancreatic islets increases glucose-dependent insulin secretion and suppresses glucagon release when glucose is elevated. Outside the pancreas, signaling in the central nervous system and gut appears to influence appetite and gastric emptying. The relative contribution of each receptor to observed clinical effects remains under investigation, and the two pathways are not simply additive in practice.
Reported outcomes in large trials include dose-dependent weight reduction and improvements in glycemic markers over periods ranging from several months to more than a year. Whether the compound alters long-term cardiovascular or renal outcomes is being examined in dedicated outcome studies, so those questions remain open. Labeling describes gastrointestinal effects such as nausea and diarrhea, which tend to appear during dose escalation. Discontinuation rates and the durability of effects after treatment stops vary across study populations and are still debated.

APCs naturally have a role in fighting tumors, via stimulation of B and cytotoxic T cells to respectively produce antibodies against tumor-related antigen and kill malignant cells. Dendritic cells, presenting tumor-specific antigen to T cells, are key to this process. Cancer therapies have included treating the patient with increased numbers of dendritic cells or cancer-specific T cells. However, newer therapies have turned to genetically engineered artificial antigen-presenting cells designed to prime the immune system to attack malignant cells. Some artificial APCs are derived from human cells; others are acellular, containing MHC proteins, co-stimulatory molecules and the necessary peptides. The APC activator IMP321 is being tested in clinical trials to accelerate the immune reaction to eliminate metastatic breast cancer or melanoma. Handheld electronics mostly use lithium polymer batteries (with a polymer gel as an electrolyte), a lithium cobalt oxide (LiCoO2) cathode material, and a graphite anode, which together offer high energy density. Lithium iron phosphate (LiFePO4), lithium manganese oxide (LiMn2O4 spinel, or Li2MnO3-based lithium-rich layered materials, LMR-NMC), and lithium nickel manganese cobalt oxide (LiNiMnCoO2 or NMC) may offer longer life and a higher discharge rate. NMC and its derivatives are widely used in the electrification of transport, one of the main technologies (combined with renewable energy) for reducing greenhouse gas emissions from vehicles. Lithium nickel cobalt aluminum oxide (NCA) is another high-energy lithium-ion chemistry commonly used in electric vehicle batteries.
==== Near-CDP ==== Continuous Data Protection (CDP) refers to a backup that instantly saves a copy of every change made to the data. This allows restoration of data to any point in time and is the most comprehensive and advanced data protection. Near-CDP backup applications—often marketed as "CDP"—automatically take incremental backups at a specific interval, for example every 15 minutes, one hour, or 24 hours. They can therefore only allow restores to an interval boundary. Near-CDP backup applications use journaling and are typically based on periodic "snapshots", read-only copies of the data frozen at a particular point in time. Near-CDP (except for Apple Time Machine) intent-logs every change on the host system, often by saving byte or block-level differences rather than file-level differences. This backup method differs from simple disk mirroring in that it enables a roll-back of the log and thus a restoration of old images of data. Intent-logging allows precautions for the consistency of live data, protecting self-consistent files but requiring applications "be quiesced and made ready for backup." Near-CDP is more practicable for ordinary personal backup applications, as opposed to true CDP, which must be run in conjunction with a virtual machine or equivalent and is therefore generally used in enterprise client-server backups.

=== Biosynthetic pathway === The metabolic pathway to produce ergothioneine starts with the methylation of histidine to produce histidine betaine (hercynine). The sulfur atom is then incorporated from cysteine. The biosynthetic genes of ergothioneine have been described in detail for Mycobacterium smegmatis, Neurospora crassa, Schizosaccharomyces pombe (with homologues in Aspergillus, a genus important in food fermentation), and Caldithrix abyssi. This pathway has recently also been discovered in plants. Different groups of organisms use different approaches to sulfur-addition. Aerobic bacteria and fungi use an O2-dependent reaction that is catalyzed by a mononuclear non-heme iron enzyme, with cysteine or γ-glutamylcysteine as the sulfur source. Green sulfur bacteria and some archaea use a rhodanese-like sulfur transferase to perform oxidative polar substitution. Caldithrix uses a metallopterin-dependent bifunctional enzyme that combines an N-terminal domain similar to a tungsten-dependent acetylene hydratase and a C-terminal cysteine desulfurase domain. Homologs of the Caldithrix system are found in anaerobic bacteria and some archaea.
Here, the resulting dimer has a stoichiometry different from the initial pair of monomers. Disaccharides need not be composed of the same monosaccharides to be considered dimers. An example is sucrose, a dimer of fructose and glucose, which follows the same reaction equation as presented above. Amino acids can also form dimers, which are called dipeptides. An example is glycylglycine, consisting of two glycine molecules joined by a peptide bond. Other examples include aspartame and carnosine.

Members of the Ugandan Parliament commended the company for pioneering drug development in underrepresented areas such as oncology and vaccine production.
=== Cross-activity === Epitopes are sometimes cross-reactive. This property is exploited by the immune system in regulation by anti-idiotypic antibodies (originally proposed by Nobel laureate Niels Kaj Jerne). If an antibody binds to an antigen's epitope, the paratope could become the epitope for another antibody that will then bind to it. If this second antibody is of IgM class, its binding can upregulate the immune response; if the second antibody is of IgG class, its binding can downregulate the immune response.

=== Military archives === The Defence Historical Service (SHD), located at the Château de Vincennes, preserves military personnel files. Many of these files contain copies, sometimes complete, of Parisian civil records. The online databases include name indexes for officer career records. On-site, one can consult muster rolls for all army units, which record each soldier’s date and place of birth, residence, and the names of parents and spouses. The Paris Archives also retain documents on military recruitment in the Seine department, including alphabetical tables (1872–1940) and muster rolls (1887–1921). These archives are accessible online.
Electrostatic interaction: In an aqueous environment, the oppositely charged groups in amino acid side chains within the active site and substrates attract each other, which is termed electrostatic interaction. For example, when a carboxylic acid (R-COOH) dissociates into RCOO− and H+ ions, COO− will attract positively charged groups such as protonated guanidine side chain of arginine. Hydrogen bond: A hydrogen bond is a specific type of dipole-dipole interaction between a partially positive hydrogen atom and a partially negative electron donor that contain a pair of electrons such as oxygen, fluorine and nitrogen. The strength of hydrogen bond depends on the chemical nature and geometric arrangement of each group. Van der Waals force: Van der Waals force is formed between oppositely charged groups due to transient uneven electron distribution in each group. If all electrons are concentrated at one pole of the group this end will be negative, while the other end will be positive. Although the individual force is weak, as the total number of interactions between the active site and substrate is massive the sum of them will be significant. Hydrophobic interaction: Non-polar hydrophobic groups tend to aggregate together in the aqueous environment and try to leave from polar solvent. These hydrophobic groups usually have long carbon chain and do not react with water molecules. When dissolving in water a protein molecule will curl up into a ball-like shape, leaving hydrophilic groups in outside while hydrophobic groups are deeply buried within the centre.
== Substrate specificity == Serine proteases are characterised by a distinctive structure, consisting of two beta-barrel domains that converge at the catalytic active site. These enzymes can be further categorised based on their substrate specificity as either trypsin-like, chymotrypsin-like or elastase-like.
These occur at asparagine (Asn) amino acid numbers 96, 135, 155, and 192 in humans and at similar amino acid numbers in other species. All these sites are occupied by covalently attached oligosaccharide side-chains in the predominant form of human antithrombin, α-antithrombin, resulting in a molecular weight for this form of antithrombin of 58,200. The potential glycosylation site at asparagine 135 is not occupied in a minor form (around 10%) of antithrombin, β-antithrombin (see Figure 1). Recombinant antithrombins with properties similar to those of normal human antithrombin have been produced using baculovirus-infected insect cells and mammalian cell lines grown in cell culture. These recombinant antithrombins generally have different glycosylation patterns to normal antithrombin and are typically used in antithrombin structural studies. For this reason many of the antithrombin structures stored in the protein data bank and presented in this article show variable glycosylation patterns. Antithrombin begins in its native state, which has a higher free energy compared to the latent state, which it decays to on average after 3 days. The latent state has the same form as the activated state – that is, when it is inhibiting thrombin.
=== Agriculture === The primary use of ammonium sulfate is as a fertilizer for alkaline soils. In the soil, the ammonium ion is released and forms a small amount of acid, lowering the pH balance of the soil, while contributing essential nitrogen for plant growth. One disadvantage to the use of ammonium sulfate is its low nitrogen content relative to ammonium nitrate or urea, which elevates transportation costs. It is also used as an agricultural spray adjuvant for water-soluble insecticides, herbicides, and fungicides. There, it functions to bind iron and calcium cations that are present in both well water and plant cells. It is particularly effective as an adjuvant for 2,4-D (amine), glyphosate, and glufosinate herbicides.

It is a synthetic peptide of 39 amino acids bearing a lipid side chain. Its size and architecture place it outside the small-molecule class, and laboratories generally handle it with the precautions used for biologic-like molecules.
The fatty diacid side chain promotes strong binding to serum albumin, which slows clearance and yields a half-life of roughly five days. That profile supports once-weekly administration in clinical use.
In vitro assays detect activity at both the GIP and GLP-1 receptors, but the activity ratio depends on the assay system and the signalling pathway measured. The relative contribution of each receptor to clinical effects is still being characterised.
It activates both the glucose-dependent insulinotropic polypeptide receptor and the glucagon-like peptide-1 receptor. This dual activity separates it from agents that act on only one of the two receptors. The relative contribution of each receptor to clinical effects remains an open area of study.
