The compound first appeared in the scientific literature as an investigational agent for type 2 diabetes. Clinical development proceeded through phase 1, phase 2, and phase 3 programs that measured glycemic control as a primary endpoint while recording body weight as a secondary outcome. Regulatory approval in the United States followed in 2022 for glycemic control, and a separate indication for chronic weight management was added later. Subsequent trials have examined cardiovascular outcomes in adults with elevated cardiovascular risk. Debates continue over how much of the observed effect derives from each receptor arm.
Structural work on the molecule centers on a C20 fatty diacid moiety attached through a linker to the peptide backbone. This side chain promotes reversible binding to serum albumin, which slows renal clearance and supports a prolonged action profile. The peptide backbone incorporates aminoisobutyric acid substitutions that limit recognition by digestive enzymes. Together these modifications produce a molecule that is stable enough for subcutaneous delivery but still dependent on careful manufacturing control. Analytical characterization of the active pharmaceutical ingredient typically follows the conventions used for other synthetic peptides.
Published work supports the view that engaging two incretin receptors produces changes in glucose handling and body weight larger than those seen with single-receptor activation. Why that difference arises is not fully settled. Open questions include how much of the observed weight effect depends on central versus peripheral signaling, and whether the two receptors form interacting complexes. Most reported findings come from controlled trials and animal models, and translation between species is imperfect. Further research is expected to refine these points over time.
Tirzepatide is a synthetic peptide built from 39 amino acid residues. Its sequence is related to human glucose-dependent insulinotropic polypeptide, with modifications that include a C-terminal extension and a C20 fatty diacid joined through a linker. Those changes raise the molecule's affinity for serum albumin, which slows renal filtration and lengthens the time it stays in circulation. The free base has an average molecular mass near 4813.5 daltons. The compound is made by solid-phase peptide synthesis followed by chromatographic purification.

The peptide backbone contains 39 amino acids and includes alpha-aminoisobutyric acid residues, which are not among the standard proteinogenic set. A C20 fatty diacid moiety is attached through a linker, allowing the compound to bind serum albumin and extend its circulation time. This albumin binding is the main reason the molecule supports once-weekly administration rather than more frequent dosing. The measured molecular mass is approximately 4,813 daltons, placing it firmly in the peptide rather than small-molecule class.
Tirzepatide is a synthetic peptide that activates both the glucose-dependent insulinotropic polypeptide (GIP) and glucagon-like peptide-1 (GLP-1) receptors. This dual agonist profile distinguishes it from earlier incretin-based compounds that act on a single receptor. The molecule was engineered from the native GIP sequence and carries several non-natural residues that slow enzymatic breakdown. Researchers designed it to combine the insulinotropic effects of GIP signaling with the appetite and gastric-emptying effects associated with GLP-1 activation.
The peptide shares degradation routes common to modified peptides: deamidation of asparagine and glutamine residues, oxidation of methionine, and backbone hydrolysis under extreme pH. Lyophilized material is generally more stable than a solution, and residual water content directly affects the rate of hydrolysis. In liquid form, aggregation and visible particles can appear after agitation or repeated freeze-thaw cycles. Stability studies therefore track monomer content, aggregate content, and potency over months under defined temperature and humidity.
Cold-chain handling is standard for formulated product, with dry powder stored frozen and ready-to-use solutions refrigerated. Light exposure is minimized because photodegradation of certain amino acid side chains is possible. Shipping and temperature-excursion studies are used to establish whether short deviations affect quality attributes. Documentation supplied with research material usually includes a certificate of analysis listing purity, identity confirmation, and water or residual solvent content. Users are expected to confirm that material meets the stated specification before use.
Identity and purity of tirzepatide are assessed mainly by reversed-phase high-performance liquid chromatography with ultraviolet detection, often paired with mass spectrometry. Because the molecule carries several modifications, gradient conditions are adjusted to resolve the intact peptide from deamidation and oxidation products. Enzymatic digestion followed by peptide mapping confirms the primary sequence and locates specific modifications. Quantitation in biological matrices typically uses liquid chromatography with tandem mass spectrometry after solid-phase extraction. Immunoassays are used less often, since antibody cross-reactivity with closely related peptides can bias results.
Degradation pathways for tirzepatide include deamidation, oxidation, and aggregation, which are common for therapeutic peptides. These processes can be monitored by size-exclusion chromatography (SEC) for aggregates and ion-exchange chromatography for charge variants. Forced degradation studies under acidic, basic, oxidative, and thermal stress help identify potential impurities. The exact stability profile depends on formulation, concentration, and container-closure system.
Analytical characterization of tirzepatide typically employs reversed-phase high-performance liquid chromatography (RP-HPLC) for purity assessment and peptide mapping. Mass spectrometry, often coupled with electrospray ionization, confirms molecular weight and sequence integrity. Amino acid analysis and capillary electrophoresis may also be used to detect impurities or degradation products. These methods are essential for batch release and stability studies.
=== Nanomaterials === Nanomaterials include carbon-based materials, metal oxides, metals, and quantum dots. Nanomaterials can enter the environment during their manufacturing, consumer use, or disposal. Due to their small size, nanomaterials behave differently than larger particles. They have a high surface area to volume ratio, which can lead to increased reactivity and the potential to transport throughout the environment. Nanomaterials are challenging to detect and monitor due to their size and the absence of standardized methods for measuring their presence and concentration in various media.
Two pharmacological agents intended to prevent scarring in mild idiopathic fibrosis are pirfenidone, which reduced reductions in the 1-year rate of decline in FVC and reduced the decline in distances on the 6-minute walk test, but had no effect on respiratory symptoms, and is nintedanib, which acts as an antifibrotic, mediated through the inhibition of a variety of tyrosine kinase receptors (including platelet-derived growth factor, fibroblast growth factor, and vascular endothelial growth factor). A randomized clinical trial showed it reduced lung-function decline and acute exacerbations. Preclinical studies on monoclonal antibodies that target the pulmonary endothelial aggregation receptor 1 (PEAR1) (which plays a crucial role in fibroblast activation and the progression of fibrosis) have shown promising results in slowing disease progression and improving lung function. Anti-inflammatory agents have only limited success in reducing the fibrotic process. Some other types of fibrosis, such as non-specific interstitial pneumonia, may respond to immunosuppressive therapy such as corticosteroids. But only a minority of patients respond to corticosteroids alone, so additional immunosuppressants, such as cyclophosphamide, azathioprine, methotrexate, penicillamine, and cyclosporine may be used. Colchicine has also been used with limited success. Nerandomilast (Jascayd) was approved for medical use in the United States in October 2025.
== Composition == In order to evaluate Durovic's and Ivy's claims, the National Cancer Institute (NCI) requested samples of Krebiozen. These were provided on two occasions to the NCI and one occasion to the U.S. Food and Drug Administration (FDA). Durovic proposed to charge the NCI $170,000 per gram of Krebiozen, although ultimately the samples were provided free of charge. However, the FDA spectrophotometry team, led by Alma Levant Hayden, proved the samples consisted only of creatine (as the monohydrate), a simple and inexpensive amino acid, dissolved in white mineral oil. When the FDA went back to test previously acquired samples of Krebiozen, they found that many lacked even the creatine monohydrate, and consisted solely of white mineral oil.
When the band emerged yet again in 1993, Hartman and Porter had retired from the road for good but Knudsen and McFee rejoined the Doobie Brothers on a full-time basis after Southern Pacific disbanded. Joined by Ockerman, Bumpus and Weeks, the group toured with Four Wheel Drive as the opening act. After Weeks left the tour to resume his session work, Cowan played bass for both bands. Bumpus also left to join the reunited Steely Dan, giving way to saxophonist, keyboardist and harmonica player Danny Hull. Former band member Chet McCracken temporarily filled in for an injured Hossack in July 1993. Their 1994 tour included co-headlining appearances with Foreigner. With renewed energy in the mid-1990s, the band began to experiment with different arrangements of several tunes. They even pulled from McDonald's songbook from time to time, eventually restoring "Takin' it to the Streets" to the setlist with Simmons and new bass guitarist Skylark (who joined in 1995) substituting for McDonald on lead vocals.
In 1912, Frederick Gowland Hopkins demonstrated that unknown accessory factors found in milk, other than carbohydrates, proteins, and fats were necessary for growth in rats. Hopkins received a Nobel Prize for this discovery in 1929. One year later, Elmer McCollum, a biochemist at the University of Wisconsin–Madison, and colleague Marguerite Davis identified a fat-soluble nutrient in butterfat and cod liver oil. Their work confirmed that of Thomas Burr Osborne and Lafayette Mendel, at Yale, also in 1913, which suggested a fat-soluble nutrient in butterfat. The "accessory factors" were termed "fat-soluble" in 1918 and later "vitamin A" in 1920. In 1931, Swiss chemist Paul Karrer described the chemical structure of vitamin A. Retinoic acid and retinol were first synthesized in 1946 and 1947 by two Dutch chemists, David Adriaan van Dorp and Jozef Ferdinand Arens. In 1967, George Wald was a co-recipient of the Nobel Prize in Physiology and Medicine "..."for their discoveries concerning the primary physiological and chemical visual processes in the eye." Photoreceptor cells in the eye contain a chromophore composed of the protein opsin and 11-cis retinal. When struck by light, 11-cis retinal undergoes photoisomerization to all-trans retinal and via signal transduction cascade sends a nerve signal to the brain. The all-trans retinal is reduced to all-trans retinol and travels back to the retinal pigment epithelium to be recycled to 11-cis retinal and conjugated to opsin.

Diversity of an individual's self-antigen presentation, mediated by MHC self-antigens, is attained in at least three ways: (1) an organism's MHC repertoire is polygenic (via multiple, interacting genes); (2) MHC expression is codominant (from both sets of inherited alleles); (3) MHC gene variants are highly polymorphic (diversely varying from organism to organism within a species). Sexual selection has been observed in male mice choosing to mate with females with different MHCs. Also, at least for MHC I presentation, there has been evidence of antigenic peptide splicing, which can combine peptides from different proteins, vastly increasing antigen diversity.
Trichilia catigua is a flowering plant species in the genus Trichilia. The species is used in folk medicine and shamanism in the aphrodisiac and stimulant catuaba. Cinchonain-Ib is a flavonolignan found in the bark of T. catigua. Catuaba from Trichilia catigua was found to increase the release of Serotonin as well as Dopamine in rats. Additionally, Trichilia catigua was shown to have neuroprotective effects in rats.
== Early life and education == Shaw was born in South Boston, and also grew up in Brookline, Massachusetts. Her mother was from South Boston. Shaw's parents divorced when she was four years old. She grew up in a single-mother home with an older half-brother, who owns a bar in Boston. Shaw attended Michael Driscoll School and Brookline High School. At Michael Driscoll, Shaw played full court streetball at a nearby neighborhood park. Shaw said that basketball was a constant of her childhood, and she incorporated it into the pilot and third episode of SMILF. After receiving a scholarship in her junior year, Shaw transferred to the private school Milton Academy in Milton, Massachusetts, where she graduated in 2000. In 2007, she graduated from Barnard College with a degree in literature.
Plutonium is a reactive metal. In moist air or moist argon, the metal oxidizes rapidly, producing a mixture of oxides and hydrides. If the metal is exposed long enough to a limited amount of water vapor, a powdery surface coating of PuO2 is formed. Also formed is plutonium hydride but an excess of water vapor forms only PuO2. Plutonium shows enormous, and reversible, reaction rates with pure hydrogen, forming plutonium hydride. It also reacts readily with oxygen, forming PuO and PuO2 as well as intermediate oxides; plutonium oxide fills 40% more volume than plutonium metal. The metal reacts with the halogens, giving rise to compounds with the general formula PuX3 where X can be F, Cl, Br or I and PuF4 is also seen. The following oxyhalides are observed: PuOCl, PuOBr and PuOI. It will react with carbon to form PuC, nitrogen to form PuN and silicon to form PuSi2. The organometallic chemistry of plutonium complexes is typical for organoactinide species; a characteristic example of an organoplutonium compound is plutonocene. Computational chemistry methods indicate an enhanced covalent character in the plutonium-ligand bonding. Powders of plutonium, its hydrides and certain oxides like Pu2O3 are pyrophoric, meaning they can ignite spontaneously at ambient temperature and are therefore handled in an inert, dry atmosphere of nitrogen or argon. Bulk plutonium ignites only when heated above 400 °C. Pu2O3 spontaneously heats up and transforms into PuO2, which is stable in dry air, but reacts with water vapor when heated.
=== Intracellular receptors === Intracellular receptors exist freely in the cytoplasm, nucleus, or can be bound to organelles or membranes. For example, the presence of nuclear and mitochondrial receptors is well documented. The binding of a ligand to the intracellular receptor typically induces a response in the cell. Intracellular receptors often have a level of specificity, this allows the receptors to initiate certain responses when bound to a corresponding ligand. Intracellular receptors typically act on lipid soluble molecules. The receptors bind to a group of DNA binding proteins. Upon binding, the receptor-ligand complex translocates to the nucleus where they can alter patterns of gene expression. Steroid hormone receptors are found in the nucleus, cytosol, and also on the plasma membrane of target cells. They are generally intracellular receptors (typically cytoplasmic or nuclear) and initiate signal transduction for steroid hormones which lead to changes in gene expression over a time period of hours to days. The best studied steroid hormone receptors are members of the nuclear receptor subfamily 3 (NR3) that include receptors for estrogen (group NR3A) and 3-ketosteroids (group NR3C). In addition to nuclear receptors, several G protein-coupled receptors and ion channels act as cell surface receptors for certain steroid hormones.

Steps to organise two further battalions of black Southern Rhodesians were abandoned because of the conviction of the colony's overall military commander, Brigadier E R Day, that it was important "to preserve a fair balance" between black and white troops, and that raising the men would take too long in any case.
== Comorbidity == Fibromyalgia as a stand-alone diagnosis is uncommon, as most fibromyalgia patients often have other chronic overlapping pain problems, mental disorders or other medical conditions. Comorbidities may be associated with more intense pain and other symptoms. Fibromyalgia is associated with mental health issues, including anxiety, post-traumatic stress disorder (PTSD), bipolar disorder, alexithymia, and depressive disorders. Approximately one-third of patients presenting with an FM diagnosis also meet the criteria for PTSD, and are five times more likely to have major depressive disorder than the general population. The pain and limited energy of fibromyalgia often leads to less activity, leading to social isolation and increased stress levels—both of which tend to cause anxiety and depression. Separation of depression symptoms from those of fibromyalgia is difficult. Chronic pain conditions are often comorbid with fibromyalgia, including neuropathic pain, chronic tension headaches, myofascial pain syndrome, and temporomandibular joint dysfunction. Neurological disorders that have been linked to pain or fibromyalgia include multiple sclerosis, post-polio syndrome, and Parkinson's disease. Fibromyalgia may share pathogenetic overlap with other syndromes, including myalgic encephalomyelitis/chronic fatigue syndrome and irritable bowel syndrome (IBS). Fibromyalgia has been reported to occur as a comorbidity in 20–30% of individuals with rheumatic diseases, including rheumatoid arthritis (RA).
Shear-thickening (dilatant) liquids, whose viscosity increases with the rate of shear strain. Shear-thinning liquids, whose viscosity decreases with the rate of shear strain. Thixotropic liquids, that become less viscous over time when shaken, agitated, or otherwise stressed. Rheopectic liquids, that become more viscous over time when shaken, agitated, or otherwise stressed. Bingham plastics that behave as a solid at low stresses but flow as a viscous fluid at high stresses. Trouton's ratio is the ratio of extensional viscosity to shear viscosity. For a Newtonian fluid, the Trouton ratio is 3. Shear-thinning liquids are very commonly, but misleadingly, described as thixotropic. Viscosity may also depend on the fluid's physical state (temperature and pressure) and other, external, factors. For gases and other compressible fluids, it depends on temperature and varies very slowly with pressure. The viscosity of some fluids may depend on other factors. A magnetorheological fluid, for example, becomes thicker when subjected to a magnetic field, possibly to the point of behaving like a solid.

It activates both the glucose-dependent insulinotropic polypeptide receptor and the glucagon-like peptide-1 receptor. This dual activity separates it from agents that act on only one of the two receptors. The relative contribution of each receptor to clinical effects remains an open area of study.
United States approval for type 2 diabetes was granted in 2022, and a chronic weight management indication was added later. Approval timelines differ across other jurisdictions. Earlier human data came from phase 1 and phase 2 trials published before those decisions.
Native hormones are short-lived peptides that enzymes degrade within minutes. Tirzepatide carries non-natural residues and a fatty acid chain that resist this breakdown and extend circulation time. The result is a longer interval between administrations than the natural hormones could support.
It is a synthetic peptide and a dual agonist of two incretin receptors. It is not a small molecule, and it is not structurally related to the older single-receptor peptide agonists.
