The short version of GLP-1 receptor fits in a sentence. The long version — which is the one that helps — is below.
Reviewed 2026-01-28. Anything still debated is marked as such rather than presented as settled.
Both receptors are class B G protein-coupled receptors that signal largely through Gs-mediated cyclic AMP production. Activation within pancreatic islets increases glucose-dependent insulin secretion and suppresses glucagon release when glucose is elevated. Outside the pancreas, signaling in the central nervous system and gut appears to influence appetite and gastric emptying. The relative contribution of each receptor to observed clinical effects remains under investigation, and the two pathways are not simply additive in practice.
Reported outcomes in large trials include dose-dependent weight reduction and improvements in glycemic markers over periods ranging from several months to more than a year. Whether the compound alters long-term cardiovascular or renal outcomes is being examined in dedicated outcome studies, so those questions remain open. Labeling describes gastrointestinal effects such as nausea and diarrhea, which tend to appear during dose escalation. Discontinuation rates and the durability of effects after treatment stops vary across study populations and are still debated.
Tirzepatide is a synthetic peptide developed as a dual agonist at the glucose-dependent insulinotropic polypeptide and glucagon-like peptide-1 receptors. Its structure is built on a GIP-derived backbone with non-natural amino acid substitutions and a fatty diacid side chain that promotes albumin binding and slows clearance. That modification supports once-weekly subcutaneous dosing. Registrational trial programs reported reductions in body weight and glycated hemoglobin alongside the drug's glycemic effects.
Verification of research-grade material involves checking purity, sequence and counter-ion content against a certificate of analysis. Reported purity figures usually reflect chromatographic area percentage and do not by themselves establish biological activity. Independent laboratories may repeat mass confirmation and peptide mapping to detect substitutions or truncations. Open questions concern how residual solvents, trace metals and subtle conformational variants affect measured behavior, and how consistently different suppliers define their specifications. Documentation of analytical methods matters as much as the headline purity number when results are compared across studies.
Routine characterization relies on reversed-phase high-performance liquid chromatography, often coupled to mass spectrometry, to confirm identity and estimate purity. Peptide mapping after enzymatic digestion verifies the amino acid sequence and locates appended groups such as the fatty acid chain. Size-exclusion chromatography detects aggregates and fragments, while ion-exchange chromatography resolves charge variants. Circular dichroism and nuclear magnetic resonance supply secondary and higher-order structural information in research settings. No single technique covers every attribute, so laboratories combine orthogonal methods and compare outcomes against a reference standard where one exists.

Identity and purity are assessed by reversed-phase high-performance liquid chromatography, with mass confirmation by electrospray ionisation mass spectrometry. Peptide mapping after enzymatic digestion verifies the primary sequence. Size-exclusion chromatography quantifies aggregates, while circular dichroism provides a secondary-structure fingerprint. Bioanalytical quantification in plasma uses immunoassay or LC-MS/MS. Reported purity for research-grade lots is commonly 95 percent or higher, and residual water content is checked by Karl Fischer titration.
As a peptide, tirzepatide is handled as a lyophilised solid in research settings and as a preserved solution in finished products. Aqueous solubility is pH dependent and reaches a minimum near the isoelectric point, which lies close to pH 5.4. Stock solutions are typically prepared in neutral or slightly basic buffer to limit precipitation. The solid is hygroscopic and should be equilibrated to room temperature before opening so that condensation does not form on the powder surface.
Cold-chain handling is standard for formulated product, with dry powder stored frozen and ready-to-use solutions refrigerated. Light exposure is minimized because photodegradation of certain amino acid side chains is possible. Shipping and temperature-excursion studies are used to establish whether short deviations affect quality attributes. Documentation supplied with research material usually includes a certificate of analysis listing purity, identity confirmation, and water or residual solvent content. Users are expected to confirm that material meets the stated specification before use.
Identity and purity of tirzepatide are assessed mainly by reversed-phase high-performance liquid chromatography with ultraviolet detection, often paired with mass spectrometry. Because the molecule carries several modifications, gradient conditions are adjusted to resolve the intact peptide from deamidation and oxidation products. Enzymatic digestion followed by peptide mapping confirms the primary sequence and locates specific modifications. Quantitation in biological matrices typically uses liquid chromatography with tandem mass spectrometry after solid-phase extraction. Immunoassays are used less often, since antibody cross-reactivity with closely related peptides can bias results.
The peptide shares degradation routes common to modified peptides: deamidation of asparagine and glutamine residues, oxidation of methionine, and backbone hydrolysis under extreme pH. Lyophilized material is generally more stable than a solution, and residual water content directly affects the rate of hydrolysis. In liquid form, aggregation and visible particles can appear after agitation or repeated freeze-thaw cycles. Stability studies therefore track monomer content, aggregate content, and potency over months under defined temperature and humidity.
Published work supports the view that engaging two incretin receptors produces changes in glucose handling and body weight larger than those seen with single-receptor activation. Why that difference arises is not fully settled. Open questions include how much of the observed weight effect depends on central versus peripheral signaling, and whether the two receptors form interacting complexes. Most reported findings come from controlled trials and animal models, and translation between species is imperfect. Further research is expected to refine these points over time.
Tirzepatide is a synthetic peptide built from 39 amino acid residues. Its sequence is related to human glucose-dependent insulinotropic polypeptide, with modifications that include a C-terminal extension and a C20 fatty diacid joined through a linker. Those changes raise the molecule's affinity for serum albumin, which slows renal filtration and lengthens the time it stays in circulation. The free base has an average molecular mass near 4813.5 daltons. The compound is made by solid-phase peptide synthesis followed by chromatographic purification.
At the receptor level, tirzepatide activates both the glucose-dependent insulinotropic polypeptide receptor and the glucagon-like peptide-1 receptor. Both belong to the class B family of G protein-coupled receptors and signal largely through cyclic AMP accumulation. The compound binds the two receptors with differing affinity, and the pattern of signaling at each site is described in the literature as biased rather than simply proportional to occupancy. Tissues carrying these receptors include pancreatic islets, adipose tissue, the central nervous system, and the gastrointestinal tract. The relative weight of each receptor population in producing metabolic effects continues to be studied.
An extended fatty diacid moiety promotes binding to serum albumin, which slows renal clearance and extends the circulating half-life to roughly five days. That property supports once-weekly administration and largely explains the dosing interval described in clinical reports. Published data come mainly from large randomised programmes that evaluated glycaemic control and body weight over periods of many months. Long-term outcomes beyond those trial windows, including what happens after treatment stops, remain an active area of investigation.
Tirzepatide is a synthetic peptide built from thirty-nine amino acids. Its sequence is derived from native glucose-dependent insulinotropic polypeptide, or GIP, with several non-natural residues and a fatty diacid side chain attached through a linker. The molecule behaves as a dual agonist at two incretin receptors, GIP and GLP-1, instead of targeting a single receptor. This dual engagement separates it from earlier single-receptor incretin compounds and underpins most of its reported pharmacological activity.
At the receptor level, the compound binds both GIP and GLP-1 receptors and triggers downstream signalling that raises cyclic AMP in target cells. GLP-1 receptor activation is associated with glucose-dependent insulin release, slower gastric emptying, and reduced appetite signalling. GIP receptor activation contributes effects that are less completely characterised, and how much each receptor adds to the overall clinical response is still an open question. The two pathways appear to interact in a complementary rather than a purely additive way.
In order to excite surface plasmon polaritons in a resonant manner, one can use electron bombardment or incident light beam (visible and infrared are typical). The incoming beam has to match its momentum to that of the plasmon. In the case of p-polarized light (polarization occurs parallel to the plane of incidence), this is possible by passing the light through a block of glass to increase the wavenumber (and the momentum), and achieve the resonance at a given wavelength and angle. S-polarized light (polarization occurs perpendicular to the plane of incidence) cannot excite electronic surface plasmons. Electronic and magnetic surface plasmons obey the following dispersion relation:
The OSA frequency increase with age for the women, and is in particular linked to the onset of menopause and associated hormonal changes. The mortality is higher for women. If studied carefully in a sleep lab by polysomnography (formal "sleep study"), it is believed that approximately 1 in 5 American adults would have at least mild OSA. In the US, some studies report that it is more frequent among the Hispanic and African American population than among the white population.
=== Pharmacokinetics === The oral activity of nandrolone has been studied. With oral administration of nandrolone in rodents, it had about one-tenth of the potency of subcutaneous injection of nandrolone. Nandrolone has very low affinity for human serum sex hormone-binding globulin (SHBG), about 5% of that of testosterone and 1% of that of DHT. It is metabolized by the enzyme 5α-reductase, among others. Nandrolone is less susceptible to metabolism by 5α-reductase and 17β-hydroxysteroid dehydrogenase than testosterone. This results in it being transformed less in so-called "androgenic" tissues like the skin, hair follicles, and prostate gland and in the kidneys, respectively. Metabolites of nandrolone include 5α-dihydronandrolone, 19-norandrosterone, and 19-noretiocholanolone, and these metabolites may be detected in urine. Single intramuscular injections of 100 mg nandrolone phenylpropionate or nandrolone decanoate have been found to produce an anabolic effect for 10 to 14 days and 20 to 25 days, respectively. Conversely, unesterified nandrolone has been used by intramuscular injection once daily.

=== Pharmacodynamics === Vosilasarm is selective androgen receptor modulator (SARM), or a tissue-selective mixed agonist or partial agonist of the androgen receptor (AR). This receptor is the biological target of endogenous androgens like testosterone and dihydrotestosterone (DHT) and of synthetic anabolic steroids like nandrolone and oxandrolone. Vosilasarm shows high affinity for the AR, with a Ki value of 7 nM (relative to 29 nM for testosterone and 10 nM for DHT). It shows good selectivity for the AR over other steroid hormone receptors, with the closest off-target receptor being the progesterone receptor (IC50 = 750 nM versus 0.2 nM for progesterone). Vosilasarm also shows potent efficacy in terms of AR activation, with an EC50 value of 0.1 nM in the C2C12 osteoblast differentiation assay. The AR is widely expressed in tissues throughout the body, including in the prostate gland, seminal vesicles, genitals, gonads, skin, hair follicles, muscle, bone, heart, adrenal cortex, liver, kidneys, and brain, among others. Vosilasarm has been found to have varying full agonist and partial agonist AR-mediated effects in different tissues, including potent agonistic and anabolic activity in muscle and bone, potent agonistic effects in AR-expressing human breast cancer cell lines, and partial agonist or antagonist activity in the prostate gland and seminal vesicles.

== Bismuth-213 == Bismuth-213 (213Bi) has a half-life of 45.6 minutes and decays mainly by beta emission to polonium-213; with only 2.1% going via alpha emission to thallium-209; however, as the polonium instantly decays by alpha, one alpha particle is emitted per atom. The amounts needed for medical use are always produced through its decay chain (the neptunium series) from either thorium-229 (limited supply due to the long life of that isotope) or actinium-225, which can be produced directly from radium-226, for example by bombardment with bremsstrahlung photons from a linear particle accelerator, knocking out a neutron and through beta decay giving actinium-225. In 1997, an antibody conjugate with 213Bi was used to treat patients with leukemia, and this isotope has otherwise been used in targeted alpha therapy (TAT) to treat a variety of cancers. Bismuth-213 is also produced in the decay of uranium-233, the fuel bred by thorium reactors, but as mentioned this goes through the long-lived thorium-229, so the production rates from each reactor will not be large.
==== Third tour 21 April to 3 June 1953 ==== In April 1953, Thomas returned alone for a third tour of America. He performed a "work in progress" version of Under Milk Wood, solo, for the first time at Harvard University on 3 May. A week later, the work was performed with a full cast at the Poetry Centre in New York. He met the deadline only after being locked in a room by Brinnin's assistant, Liz Reitell, and he was still editing the script on the afternoon of the performance; its last lines were handed to the actors as they were putting on their makeup. During this penultimate tour, Thomas met the composer Igor Stravinsky who had become an admirer after having been introduced to his poetry by W. H. Auden. They had discussions about collaborating on a "musical theatrical work" for which Thomas would provide the libretto on the theme of "the rediscovery of love and language in what might be left after the world after the bomb." The letters Stravinsky sent to Thomas during this period testify not only to the composer's sustained interest in bringing the projected opera to fruition but also to the friendship and affection that developed between them, which in turn fueled their shared excitement about a project they envisioned as both artistically ambitious and urgently attuned to the anxieties of their time.(The Opera that Never Was) The shock of Thomas's death later in the year moved Stravinsky to compose his In Memoriam Dylan Thomas for tenor, string quartet and four trombones.
=== Acting === Director Lal Jose noticed Kallingal on a magazine cover and approached her for a goat herder role in the Tamil film Mazhai Varappoguthu. The project did not go ahead, but director Shyamaprasad subsequently cast her in the lead female role of Varsha in Ritu (2009), which marked her acting debut. In 2009 she also appeared in the anthology film Kerala Cafe, in the segment Mrityunjayam, and in Neelathaamara directed by Lal Jose. In 2012, Kallingal was cast opposite Fahadh Faasil in 22 Female Kottayam, directed by Aashiq Abu, playing a nurse and rape survivor seeking justice. The film performed well commercially and won Kallingal the Kerala State Film Award for Best Actress and the Filmfare Award for Best Actress – Malayalam. The film also drew praise from critics; Sify wrote that Kallingal "poured her heart into her character" and delivered "a spectacular performance". In 2013, she hosted the Mazhavil Manorama reality show Midukki, following which the Kerala Film Chamber imposed a temporary ban on her for disregarding a recent notice by the chamber; the ban was subsequently revoked. That same year she played a housewife in August Club. Paresh C. Palicha of Rediff.com noted that Kallingal portrayed a character "who has learnt to stifle her feelings and finding other ways to give them an outlet". In 2015, she appeared alongside Manju Warrier in Rani Padmini, playing a disruptive North Indian woman on a road trip from Kerala to Shimla. The film was a commercial success and critic Deepa Soman described her performance as a "knockout".
DCPIP can also be used as an indicator for vitamin C. If vitamin C, which is a good reducing agent, is present, the blue dye, which turns pink in acidic conditions, is reduced to a colorless compound by ascorbic acid. This reaction is a redox reaction: vitamin C (ascorbic acid) is oxidized to dehydroascorbic acid, and DCPIP is reduced to the colorless compound DCPIPH2
Meropenem, sold under the brand name Merrem among others, is an intravenous carbapenem antibiotic used to treat a variety of bacterial infections. Some of these include meningitis, intra-abdominal infection, pneumonia, sepsis, and anthrax. Common side effects include nausea, diarrhea, constipation, headache, rash, and pain at the site of injection. Serious side effects include Clostridioides difficile infection, seizures, and allergic reactions including anaphylaxis. Those who are allergic to other β-lactam antibiotics are more likely to be allergic to meropenem as well. Use in pregnancy appears to be safe. It is in the carbapenem family of medications. Meropenem usually results in bacterial death through blocking their ability to make a cell wall. It is resistant to breakdown by many kinds of β-lactamase enzymes, produced by bacteria to protect themselves from antibiotics. Meropenem was patented in 1983. It was approved for medical use in the United States in 1996. It is on the World Health Organization's List of Essential Medicines. The World Health Organization classifies meropenem as critically important for human medicine.

== Childhood and education == Joseph Fruchtgarten was born in Częstochowa, Poland; his father Shama Nuta (Charles) Fruchtgarten was a grain merchant, and his mother Ella (Aisenstadt) Fruchtgarten was a French teacher. Like many other Polish Jews, the Fruchtgartens immigrated to the United States shortly before the outbreak of World War I. They lived in New York City from 1913 to 1917, and in April 1917 they moved to Minsk (then occupied by the Red Army in the midst of the Russian Civil War). Between 1917 and 1923, Fruchtgarten attended school intermittently, moving from Minsk to Siedlce to Warsaw to Berlin, and learning French, German and Latin (in addition to Polish and English). In 1923, the Fruchtgartens returned to New York and changed their name to Fruton to avoid being targets of anti-Semitism. Joseph Fruton followed his father in rejecting religion, but learned early on "not to advertise either [his] Jewishness or [his] atheism." After a few months at De Witt Clinton High School, Fruton joined the first class of students at James Madison High School. He graduated in 1927, excelling particularly in chemistry. He applied to Columbia University, and after an initial rejection—possibly because he was only 15 at the time, possibly because the school had already admitted the quota of New York Jews—his mother convinced an admissions official to reverse the decision. Inspired by the character Max Gottlieb from the Sinclair Lewis novel Arrowsmith, Fruton planned his Columbia education around becoming a scientist. The lectures and lab-work of organic chemist John M.
Trigger finger, also known as stenosing tenosynovitis, is a disorder characterized by catching or locking of the involved finger in full or near full flexion, typically when the hand is closed with force. There may be tenderness in the palm of the hand near the last skin crease (distal palmar crease). The ring finger and thumb are the most common digits. The problem is generally idiopathic (no known cause). People with diabetes might be relatively prone to trigger finger. The pathophysiology is enlargement of the flexor tendon and the A1 pulley of the tendon sheath. While often referred to as a type of stenosing tenosynovitis (which implies inflammation) the pathology is mucoid degeneration. Mucoid degeneration refers to changes in fibrous tissue, such as tendon, to have less organized collagen, more abundant extracellular matrix, and changes in the cells (fibrocytes) so as to act and look more like cartilage cells (chondroid metaplasia). Diagnosis is typically based on symptoms and signs after excluding other possible causes. Trigger digits can resolve without treatment. Treatment options that are disease modifying include steroid injections and surgery. Splinting immobilization of the finger may or may not be disease modifying.
Benzodiazepines are generally viewed as safe and effective for short-term use of two to four weeks, although cognitive impairment and paradoxical effects such as aggression or behavioral disinhibition can occur. According to the Government of Victoria's (Australia) Department of Health, long-term use can cause "impaired thinking or memory loss, anxiety and depression, irritability, paranoia, aggression, etc." A minority of people have paradoxical reactions after taking benzodiazepines such as worsened agitation or panic. Benzodiazepines are often prescribed for as-needed use, which is under-studied, but is most likely, probably safe and effective to the extent that it involves intermittent short-term use. Benzodiazepines are associated with an increased risk of suicide due to aggression, impulsivity, and negative withdrawal effects. Long-term use is controversial because of concerns about decreasing effectiveness, physical dependence, benzodiazepine withdrawal syndrome, and an increased risk of dementia and cancer. The elderly are at an increased risk of both short- and long-term adverse effects, and as a result, all benzodiazepines are listed in the Beers List of inappropriate medications for older adults. There is controversy concerning the safety of benzodiazepines during pregnancy. While they are not major teratogens, uncertainty remains as to whether they cause cleft palate in a small number of babies and whether neurobehavioural effects occur as a result of prenatal exposure; they are known to cause withdrawal symptoms in the newborn.
== Contraindications == Voriconazole is toxic to the fetus, and thus not used in pregnant women, or women who may become pregnant. People with hereditary intolerance for galactose, Lapp lactase deficiency, or glucose-galactose malabsorption should not take this drug. It should be used cautiously in people with arrhythmias or a long QTc interval. No dose adjustment is necessary for renal impairment or advanced age, but children seem to clear voriconazole faster than adults, and drug levels may need monitoring.
== Stability == In 2016, laboratory research established and compared the radiosensitivity of three organic food colorants including curcumin, carmine, and annatto to create data to be used for application whenever food products containing these food colors were to undergo the radiation process. The researchers used spectrophotometry and capillary electrophoresis to establish radiosensitivity of the three organic food colorants. Carmine samples were quite stable against radiation treatment, annatto showed limited stability, and curcumin was found to be unstable, particularly when diluted.

It acts as an agonist at both the GIP and GLP-1 receptors, two related class B G protein-coupled receptors. This dual activity distinguishes it from single-receptor GLP-1 agonists. The clinical consequences of engaging both receptors are still being characterized.
It is given by subcutaneous injection, generally on a weekly schedule. Dosing usually begins low and increases in steps to limit gastrointestinal side effects. Formulated product is supplied as a ready-to-use pen or vial in most markets.
No. Receptor engagement is documented, but how central appetite circuits, gut signals, and insulinotropic effects combine is incompletely resolved. Investigators continue to separate GIP-driven from GLP-1-driven contributions in animal and human models.
Liquid chromatography combined with mass spectrometry is the most common approach. Digestion followed by peptide mapping verifies the sequence and modification sites. Results are judged against a reference standard or a theoretically calculated mass.
